What does 5s mean here?

With diamonds agreed as trumps, you keycard and get 5d (1 or 4) response. You have all 5 and queen of trumps. What would 5s mean here? Can it ask partner to bid grand if he has SK?

Yes, but better agreements exist. I prefer for 5 to ask for specific kings ('this one or the other two'), 5NT to ask for generic extras, and 6 to ask for a third round control in clubs. That last agreement isn't particularly good, but it fits well with my meta-agreements so it is easy to remember.

Without additional discussion, I would expect 5 to ask for third round control of spades (so doubleton or Q) to bid grand.

I don't think this is practical, surely you need it to ask for K of spades. Otherwise there really isn't enough room. To get 3rd round control asks at this level, you really had to be playing kickback, so that Q ask is always lower than 5D, 5H (trump suit + 1) can be K ask, and higher can be 3rd round control asks.
